Cryopreservation of Pacific oyster ( Crassostrea gigas ) larvae: Revisiting the practical limitations and scaling up the procedure for application to hatchery ArchiMer

Pacific oyster Crassostrea gigas is one major species for aquaculture, and the development of breeding programs and the need for preservation of wild stock genetic resources prompted the need for larvae cryopreservation. The objective of the present study was to choose the most reliable protocol from several existing publications, to test its biological and practical limitations, and to adapt it to hatchery conditions. The selected protocol was characterized by a very slow freezing rate without seeding, and by the use of ethylene glycol and sucrose as cryoprotectant. Effect of age and environmental conditions on the movement characteristics of Pacific oyster (Crassostrea gigas) trochophores ArchiMer

The movement characteristics of Pacific oyster trochophores have received very little coverage in the scientific literature. Described here are not only changes in the swimming characteristics of Crassostrea gigas trochophores (size: 53-77 mu m) in relation to time after fertilization and to inter-female variation but also the effects of salinity and pH on movement characteristics. The percentage of motile trochophores was measured on images obtained through a dissecting microscope and the Velocity Average Path (VAP) was assessed using a Computer Assisted Sperm Analysis (CASA) system. Setting tools for the early assessment of the quality of thawed Pacific oyster (Crassostrea gigas) D-larvae ArchiMer

Parameters used to assess the survival of larvae after cryopreservation generally misestimate the damages that prevent larval development. The objectives of the present study were to 1) define the reliability of the survival rate, assessed at 2 and 7 days post fertilization, to estimate Pacific oyster larval quality after thawing, and 2) select complementary tools allowing an early and reliable estimation of their quality. Oyster larvae were reared for 25 h after fertilization at 19 degrees C and cryopreserved at early D-stage. Then, thawed larvae were incubated in 2-L beakers.
